چکیده

The properties of the glucose 6-phosphate-dependent forms of glycogen synthetase obtained by incubation with calcium ions or with adenosine triphosphate were compared. The two preparations showed different sensitivity to the action of heat and trypsin. In addition, the synthetase obtained with adenosine triphosphate is reconvertible to the independent form while that prepared with Ca++ is not. The possible physiological role of the two different mechanisms of formation of the glucose 6-phosphate-dependent forms of glycogen synthetase is discussed.

Glycogen Synthetase Activity in Skeletal Muscle. Interconversion of Two Forms and Control of Glycogen Synthesis.

Glycogen synthetase (uridine diphosphate glucose-glycogen glucosyltransferase, EC 2.4.1.11) has been extensively studied in liver and in skeletal muscle (3-8). In extracts from a variety of tissues, measurements of the activity of the enzymes catalyzing the steps from n-glucose 6-phosphate to glycogen suggested that glycogen synthetase was rate-limiting and, hence, was likely to be important in...

Inactivation of glycogen synthetase and activation of phosphorylase kinase by muscle adenosine 3',5'-monophosphate-dependent protein kinases.

Rabbit skeletal muscle glycogen synthetase I has been purified and obtained essentially free of phosphor-y&e, phosphorylase khmse, and glycogen synthetase kinase. Using the purified glycogen synthetase as substrate, it was determined that two separable adenosine 3’,5’-monophosphate (cyclic AMP)-dependent protein kinase fractions from skeletal muscle each catalyze the conversion of glycogen synt...

Regulation of glycogen synthetase. Specificity and stoichiometry of phosphorylation of the skeletal muscle enzyme by cyclic 3':5'-AMP-dependent protein kinase.

Complete conversion of skeletal muscle glycogen synthetase from the I form to the D form requires incorporation of 2 mol of phosphate per enzyme subunit (90,000 g). Incubation of sythetase I with low concentrations of adenosine 3':5'-monophosphate(cAMP)-dependent protein kinase (10 units/ml) and ATP (0.1 to 0.3 mM) plus magnesium acetate (10 mM) results in incorporation within 1/2 hour of 1 mol...

Characterization of glycogen synthetases and phosphorylases in transplantable rat hepatomas.

A study of the chemical and kinetic properties of a series of transplantable, chemically induced rat hepatomas has revealed profound alterations in isozyme patterns of glycogen synthetase and phosphorylase. Highly differentiated, slow-growing hepatomas retain the liver-type isozymes, but with decreased differentiation and increased growth rate these are largely or entirely replaced by nonhepati...
